Vitiligo is an autoimmune skin disease characterized by the progressive destruction of melanocytes autoreactive T cells, resulting in persistent white patches. We previously combined multiphoton microscopy (MPM) and single cell RNA sequencing (scRNA-seq) stable vitiligo lesions to identify a subset metabolically shifted keratinocytes, which we now call ‘stable keratinocytes’ (SVKs), that accumulate skin. SVKs produce high levels chemokines known induce (CXCL9, CXCL10) low Wnt signals recruit melanocytes. were associated with persistence, as they not observed acute vitiligo. also noted less numerous from patients responded punch grafting treatment compared failed respond.
